SECARA IN VITRO , WISNU ADHI SUSILA , Dr. Ir. Murwantoko, M.Si ETD Infectious diseases caused by Oomycetes become one of obstacle in aquaculture. Based on interview with some fish farmers, banana pseudostem could be used to overcome saprolegniasis. Saprolegniasis could be caused by Saprolegnia sp or Achlya sp infection. The purpose of this study were to determine the antifungal activity of metanol extract of ambon banana (Musa acuminata) pseudostem and to identify its bioactive compounds. Ambon banana pseudostem was collected fom Bantul while the infected gouramy was collected from Pingit market, Yogyakarta. The banana pseudostem was chopped, dried, and extracted. Extraction was done using maceration method with methanol as solvent. The fungi from gouramy was inoculated on potato dextrose agar (PDA) and then was incubated. Identification was done by observing its sporangium morphology. Fungi purificaton was carried out on PDA medium supplemented by ciprofloxacin 1 mg/ml. The antifungal activity assay was done using agar diffusion and dilution method. The phytochemical screening was done using 96 microwell plate and using thin layer chromatography (TLC) plates. In addition, saponin and tannin screening was also performed. The result showed the fungi which infected fish is Achlya sp. Methanol extract of ambon banana did not posses antifungal activity against Achlya sp. Phytochemistry screening showed that methanol extract ambon banana pseudostem contain terpen and saponin. Further studies was needed to determine antifungal activity from different banana species and to determine the effect of banana pseudostem application on water quality.
