الملخص:Congestive heart failure (CHF) is mainly resulted from heart fibrosis. Collagen accumulation in heart fibrosis occurs as result from activity of myofibroblast. High salt intake activates myofibroblast via angiotensin II - TGF β1 pathway. Telmisartan is a highly selective angiotensin II type 1 (AT1) receptor blocker (ARB) for treatment of hypertension and as partial agonist of PPAR-γ which decreases TGF-β1 and is predicted to prevent myocardial fibrosis. Objective To know the effect of telmisartan in variation doses related TGF-β1 and collagen volume fraction of myocardial tissue. Method Ten-week-old male Wistar Rat (n = 30) were randomized into five groups, and each group consisted of 6 rats. Group 1 : negative control. Group 2 : rats were induced by intake Nacl 8% doses 2% body weight for eight weeks. Group 3 : rats were induced by intake Nacl 8% doses 2% body weight and telmisartan 3 mg/kgBB for eight weeks. Group 4 : rats were induced by intake Nacl 8% doses 2% body weight and telmisartan 6 mg/kgBB for eight weeks. Group 5 : rats were induced by intake Nacl 8% doses 2% body weight and telmisartan 12 mg/kgBB for eight weeks. Myocardial tissue were stained with picrosirius red and collagen volume fraction were assessed by ImageJ programme. Concentration of TGF-β1 in myocardial tissue were assessed by ELISA. ANOVA test followed pos hoc test (Bonferroni test) was used to analyzed each variable. Results Concentration of TGF-β1 in group 1 (909.05 ± 14.28 pg/mL), group 2 (1360.41 ± 56.19 pg/mL), group 3 (1142.75 ± 26.50 pg/mL), group 4 (691.96 ± 34.51 pg/mL) and group 5 (435.97 ± 29.25 pg/mL). TGF- β1 significantly decreased in group 3, 4 and 5 compared in group 2, p = 0.00. Collagen volume fraction in group 1 (2.53 ± 0.11%), group 2( 19.20 ± 0.13%), group 3 (9.35 ± 0.25%), group 4 (3.97 ± 0.38%) and group 5 (2.21 ± 0.29%). Collagen volume fraction significantly decreased in group 3, 4 and 5 compared in group 2, p = 0.00. Conclusion Telmisartan decreases concentration of TGF-β1 and collagen volume fraction of myocardial tissue wistar rats induced by high salt intake.
