總結: | Vanda tricolor is one of typical orchid found in southern slope of Mount
Merapi and western slope of Mount Lawu. Different environmental condition
would affects plant growth in terms of physiological responses and anatomy of
plant. The aim of this research was to examine the physiological responses and
anatomical character of vegetative organs of V.tricolor which naturally grew on
southern slope of Mount Merapi compared to V.tricolor which naturally grew on
western slope of Mount Lawu.
Physiological parameters examined were abscisic acid level and
chlorophyll content, anatomical characters examined including roots, stem and
leaves. The association between orchid roots and mychorriza was evaluated as
well. Level of abscisic acid was determined using HPLC methode, chlorophyll
content was determined using spectrophotometry, anatomical characters was
prepared using free-hand section and non embedding using sliding mycrotome.
Distribution of mycorhizza on orchid roots were determined using triphan blue
staining. Data were analyzed with statistical analysis Independent T-test to
determine sinificantly different results. Anatomical characters also were analyzed
descriptively.
The result showed that abscisic acid level of V.tricolor grew on southern
slope of Mount Merapi higher (15,008a ppm) than V.tricolor grew on western
slope of Mount Lawu (2,978b ppm). Chlorophyll content of V.tricolor Mount
Merapi higher (0,361a mg/g) than V.tricolor Mount Lawu (0,245b mg/g).
Anatomical character showed that V.tricolor grew on southern slope of Mount
Merapi has a higher value compare to V.tricolor grew on western slope of Mount
Lawu in the thickness of velamen and exodermis of aerial roots, the trakea
number of roots, trakea diameter of stem vessel, thickness of stem bundle sheath,
thickness of mesophyll, diameter of trakea in midrib. On the other hand, V.tricolor
grew in southern slope of Mount Merapi has less value of the thickness of
periderm, the number of stem vessel, stomatal index of upper and lower
epidermis, as well as percentage of mychorriza infection compare to V.tricolor
grew in western slope of Mount Lawu. |
