Circadian variations of sulfamethoxazole pharmacokinetics were studied after a single oral administration sulfamethoxazole, 50 mg/kg bw, given at 09.00 a.m and 09.00 pm. Blood sample was taken from 0.25; 0.50; 1.00; 1.50; 2.00 ; 4.00 ; 6.00 ; 9.00; 12.00 ; 24.00 hours. Using spectrophotometer UV-VIS, the pharmacokinetics parameter was calculate for Area Under Curve (AUC), elimination rate constant (k e ), and half life (t ½). From statistical analysis it could be seen that there were significant difference on AUC 09.00 am and 21.00 pm but not in elimination rate constant (k e ) and halflife (t ½). The AUC was significantly smaller in the morning (2674.571±452.660 µg/ml.minutes) than in the nighttime (4536.532±1534.370 µg/ml.minutes) (p<0.05). The elimination rate constant (k e ) was not significantly increased in the morning (0.012±0.003/minutes) than in the night time (0.009±0.008/minutes) (p>0.05). The half-life (t 1/2 ) was not significantly shorter in the morning (34.000±2.550 minutes) than the night time (58.200±0.840 minutes) (p>0.05). Overall, the results shows the difference characteristic of pharmacokinetics due to the circadian time.
