Summary: | Electrophoresis is a biochemical method for applying polymorphic enzymes utilizable for blood individualization in forensic area. The object of this report is to choose the most appropriated electrophoretic system for typing human blood phosphoglucomutase [PGM], one of the high-potential polymorphic enzymes for blood individualization. Four electrophoretic system were compared and the results were evaluated as summarized. Cellulose acetate membranye [CAM] electrophoresis at pH 7.4 could resolve PGM isozymes in to 3 subtypes namely 2-2, 1-1, and 2-1. Agarose-starch gel polyacrylamine gel at the same pH provided the same PGM pattern with relativity lower resolution and sensitivity. Polyacrylamide gel electrophoresis [PAGE] with continuous system at pH 7.4 or with discontinous system at pH 8.9 failed to resolve the enzyme. Isoelectricfocusing [IEF] on polyacrylamide gel provided the best resolution with 4 isozymes of 10 subtypes, namely 1-, 1+, 1+1-, 2-, 2+, 2+2-, 1-2-, 1-2+, 1+2-, 1+2+, thus highly increasedthe discriminating power of PGM. Among different IEF systems tested, horizontal system of pH 5-7 and 5.3. %T provided the best solution and sensitivity. The detail of each system is clearly reported. |
