الملخص:Breast cancer is one of kind malignant disease in Indonesia and the world. The incidence and mortality is still high due to the resistance of the cancer cell, driving the efforts for discovering cancer therapeutic agent that more effectively and safely. The Gynura procumbens plants have activity as an antipyretic, anti hypotension, anti hypoglycemic and anticancer . The aim of this research was to find out the cytotoxic potency in ethyl acetate fraction of Gynura procumbens leaf to MCF - 7 and T47D breast cancer cells and to identify the active compounds that lead to cytotoxic activity. Gynura procumbens was extracted using ethanol 96%, then followed by liquid-liquid partition hexane and ethyl acetate. Ethyl acetate fraction was separated by two- dimensional TLC method to see existing patches . This method uses cellulose stationary phase with two different mobile phase is ethanol - water (1:4) and butanol - acetic acid - water ( 4:1:5 ). Furthermore, ethyl acetate fraction was separated and propagated by using preparative TLC method, the results obtained KLTP 3 subfraksi then tested the subfraksi cytotoxic activity against breast cancer cells MCF-7 and T47D by using MTT assay. The our results showed that subfraksi 1 have cytotoxic activity against MCF-7 cells with IC50 value of 605.20 µg / ml. Subfraksi 1 then separated and reproduced by preparative TLC with a mobile phase of BAW thus obtained isolates and their structures elucidated. Unpurified isolates obtained so that the compounds responsible for the cytotoxic activity of G. procumbens is unknown. Isolates were estimated to contain active compounds similar to 2- isopropyl-5-methylcyclohexanol (menthol), 2-Cyclohexyl-5-methyl acetate (acetyl menthol) and 2 - ethylhexyl Phtalate.
